چکیده

We carried out the purification of C-phycocyanin and allophycocyanin from Spirulina platensis taking advantage of the adsorption properties of the expanded beds. Initially, phycobiliproteins were released from the microalga cells by osmotic shock. Next, phycocyanins were recovered by applying the centrifuged cell suspension directly to the anion exchanger Streamline-DEAE using expanded bed columns, equilibrated with 50 mM sodium phosphate buffer, pH 7.0. After adsorption, washing was carried out in the expanded-bed mode. Having removed unbound proteins and cellular debris, the bed was allowed to sediment and phycocyanins rich solution was eluted with a downward flow of 500 mM sodium phosphate buffer, pH 7.0. Finally, we utilized conventional gel filtration and ion exchange chromatography methods for separation and purification of C-phycocyanin and allophycocyanin. The purification steps were monitored using sodium dodecyl sulfate-polyacrylamide gel electrophoresis, and the purity of recovered phycocyanins was confirmed by absorption and emission spectroscopy. The main advantage of this new method is the high yield achieved in the steps of product extraction and adsorption by expanded bed adsorption, so reducing both processing times and costs.

KINETICS AND ADSORPTION ISOTHERM OF C-PHYCOCYANIN FROM Spirulina platensis ON ION-EXCHANGE RESINS

C-phycocyanin is a natural blue dye extracted from Spirulina platensis, which has many applications in the food and pharmaceutical industries. In this paper the effect of pH and temperature on the adsorption of C-phycocyanin onto two different ion exchange resins (Streamline DEAE and Streamline Q XL) for expanded bed adsorption chromatography was investigated. Moreover, the kinetics and adsorpt...

Accelerated Solvent Extracts from Spirulina Platensis Microalga: Determination of their Antioxidant Activity and Analysis by Micellar Electrokinetic Chromatography

In this work, different extracts from the microalga Spirulina platensis are obtained using Accelerated Solvent Extraction (ASE) and four different solvents (hexane, petroleum ether, ethanol and water). Different extraction temperatures (115oC and 170oC) were tested using extraction times ranging from 9 to 15 minutes. The antioxidant activity of the different extracts is determined by means of a...

Partial Replacement of Fishmeal by Lyophylized Powder of the Microalgae Spirulina platensis in Pacific White Shrimp Diets

An eleven-week feeding trial was conducted to evaluate the growth effect of partial replacement of fishmeal by the microalga Spirulina platensis in the diet of juvenile Pacific white shrimp Litopenaeus vannamei (Boone). The results clearly indicate that Spirulina platensis constitutes an effective food ingredient for shrimp. Growth rates of a Spirulina-fed group differed highly significantly (p...
